C57BL/6 mice immunized with p60 showed antigen-specifi c T-helper I type immune response. Mouse macrophage RAW264.7 cells produced tumor necrosis factor alpha (TNF-α), interleukin-12 and interferon beta (IFN-β) in response to stimulation with recombinant p60. Administration of p60 prior to a sublethal infection with L. monocytogenes enhanced innate host resistance in naïve mice. TNF-α and IFN-β production from RAW264.7 cells and bone marrow-derived macrophages were Toll-like receptor 4 (TLR4)-dependent. The enhanced clearance of L. monocytogenes by p60 administration was not shown in C3H/HeJ mice.
