{"created":"2023-05-15T09:03:35.927514+00:00","id":4522,"links":{},"metadata":{"_buckets":{"deposit":"30ac7203-89f3-4e0a-bff0-1fc484b18a2d"},"_deposit":{"created_by":3,"id":"4522","owners":[3],"pid":{"revision_id":0,"type":"depid","value":"4522"},"status":"published"},"_oai":{"id":"oai:hirosaki.repo.nii.ac.jp:00004522","sets":["701:728:729"]},"author_link":["16855","16867","16861"],"item_8_alternative_title_22":{"attribute_name":"タイトル(ヨミ)","attribute_value_mlt":[{"subitem_alternative_title":"アンチセンス カクサン コウガク ニヨル リンゴ カジツ ノ カンジュク ヨクセイ ニカンスル ケンキュウ"}]},"item_8_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"1997-03","bibliographicIssueDateType":"Issued"},"bibliographic_titles":[{}]}]},"item_8_description_17":{"attribute_name":"フォーマット","attribute_value_mlt":[{"subitem_description":"application/pdf","subitem_description_type":"Other"}]},"item_8_description_42":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"subitem_description":"Research Paper","subitem_description_type":"Other"}]},"item_8_description_5":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"今年度は先ず、リンゴへの遺伝子導入を行うにあたり大きな要因となる、再分化系の確立を図った。栽培品種‘王林'を材料として、再分化のための培養条件を検討した。無菌的に培養したシュ-トの葉身から再分化を試みた結果、1/2MSにBAP7mg/l、NAA0.2mg/lの条件で34.4%の再分化率が得られた。また、葉身を葉柄側から葉頂にかけて2~4群の葉切片に分け、それぞれの再分化率を検討したところ、1.1~1.5cmの葉身の中央部が最も高い再分化率(75%)を示すことが明らかにされた。さらに、抗生物質の濃度の検討から形質転換体の選抜には、カナマイシン25mg/l、除菌にはセフォタキシン300mg/lが最適であることを見い出した。次に、リンゴ果実の過熟を抑制するため1-アミノシクロプロパン-1-カルボン酸(ACC)合成酵素(S-adenosyl-L-methionine methylthioadnosine-lyase,EC4.4.1.14)のcDNAを用いて、これをアンチセンスで導入し、エチレン合成系を制御出来る可能性に着目し、ACC合成酵素遺伝子のcDNAの作成を試みた。収穫期のリンゴ‘ゴ-ルデンデリシャス'の果実から全RNAを抽出し、これを鋳型としてRT-PCRを行ったところ、約1,200bpの増幅産物を得、これがACC合成酵素遺伝子のcDNAであることを確認した。今後はこのcDNAをバイナリ-ベクタ-に挿入し、リンゴ栽培種に導入する実験を開始するとともに、既にクロ-ニングされているACC合成酵素遺伝子のゲノミッククロ-ンの塩基配列の決定を進める予定である。","subitem_description_type":"Abstract"}]},"item_8_description_6":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"平成7年度~平成8年度科学研究費補助金基盤研究(C)(2)研究成果報告書,課題番号:07660003","subitem_description_type":"Other"}]},"item_8_description_7":{"attribute_name":"引用","attribute_value_mlt":[{"subitem_description":"43p ;
