ET receptor( ET-R) antagonists have recently become established as a drug essential for treating pulmonary arterial hypertension( PAH). β-arrestin was originally identified as a regulator of G-protein coupled receptor recycling, but it recently became apparent that β-arrestins act as scaffolds in their own signaling pathway. In this study, we examined the role of β-arrestin in ET-R signaling and explored its possible role in the pathogenesis of PAH.\nMethods and Results: The knockdown of β-arrestin1 or β-arrestin2 in human kidney embryonic 293 cells resulted in nhanced extracellular signal-regulated kinase (ERK) 1/2 phosphorylation in response to ET. Confocal microscopy showed that, in the absence of stimulation, transiently transfected green fluorescent protein-tagged epidermal growth factor receptors( EGFRs) were located on the plasma membrane, whereas they were internalized in response to ET, as shown by their redistribution into cellular aggregates. Pretreatment with Ro318425( a protein kinase C inhibitor) or AG1478( an EGFR antagonist) suppressed ERK1/2 phosphorylation in response to ET.\nConclusions: β-arrestins and EGFR transactivation are involved in ET-R signaling. These new insights may contribute to elucidating further layers in the pathogenesis of PAH.","subitem_description_type":"Abstract"}]},"item_3_publisher_35":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"弘前大学大学院医学研究科・弘前医学会"}]},"item_3_source_id_11":{"attribute_name":"書誌レコードID","attribute_value_mlt":[{"subitem_source_identifier":"AN00211444","subitem_source_identifier_type":"NCID"}]},"item_3_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"0439-1721","subitem_source_identifier_type":"ISSN"}]},"item_3_subject_19":{"attribute_name":"日本十進分類法","attribute_value_mlt":[{"subitem_subject":"490","subitem_subject_scheme":"NDC"}]},"item_3_text_4":{"attribute_name":"著者所属","attribute_value_mlt":[{"subitem_text_value":"Department of Cardiology, Hirosaki University Graduate School of Medicine,"}]},"item_3_version_type_18":{"attribute_name":"著者版フラグ","attribute_value_mlt":[{"subitem_version_resource":"http://purl.org/coar/version/c_970fb48d4fbd8a85","subitem_version_type":"VoR"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Narita, Noritomo"}],"nameIdentifiers":[{"nameIdentifier":"19861","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Hanada, Kenji"}],"nameIdentifiers":[{"nameIdentifier":"12011","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Senoo, Maiko"}],"nameIdentifiers":[{"nameIdentifier":"19856","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Kato, Tomo"}],"nameIdentifiers":[{"nameIdentifier":"19857","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Kudo, Natsumi"}],"nameIdentifiers":[{"nameIdentifier":"19858","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Yokono, Yoshikazu"}],"nameIdentifiers":[{"nameIdentifier":"19859","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Tsushima, Michiko"}],"nameIdentifiers":[{"nameIdentifier":"19860","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Toyama, Yuichi"}],"nameIdentifiers":[{"nameIdentifier":"19278","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Narita, Masato"}],"nameIdentifiers":[{"nameIdentifier":"19862","nameIdentifierScheme":"WEKO"}]},{"creatorNames":[{"creatorName":"Tomita, Hirofumi"}],"nameIdentifiers":[{"nameIdentifier":"11980","nameIdentifierScheme":"WEKO"},{"nameIdentifier":"100000529","nameIdentifierScheme":"研究者総覧","nameIdentifierURI":"http://hue2.jm.hirosaki-u.ac.jp/html/100000529_ja.html"}]}]},"item_files":{"attribute_name":"ファイル情報","attribute_type":"file","attribute_value_mlt":[{"accessrole":"open_date","date":[{"dateType":"Available","dateValue":"2019-04-10"}],"displaytype":"detail","filename":"HirosakiMedJ_69(1-4)_146.pdf","filesize":[{"value":"440.5 kB"}],"format":"application/pdf","licensetype":"license_note","mimetype":"application/pdf","url":{"label":"HirosakiMedJ_69(1-4)_146.pdf","url":"https://hirosaki.repo.nii.ac.jp/record/5521/files/HirosakiMedJ_69(1-4)_146.pdf"},"version_id":"1cb73a95-126c-4fed-892a-324f92c883f6"}]},"item_keyword":{"attribute_name":"キーワード","attribute_value_mlt":[{"subitem_subject":"Endothelin","subitem_subject_scheme":"Other"},{"subitem_subject":"β-arrestin","subitem_subject_scheme":"Other"},{"subitem_subject":"G-protein coupled receptor","subitem_subject_scheme":"Other"},{"subitem_subject":"Pulmonary arterial hypertension","subitem_subject_scheme":"Other"}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"Involvement of β-arrestin in Endothelin Receptor Signaling : a Possible Role in the Pathogenesis of Pulmonary Arterial Hypertension","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"Involvement of β-arrestin in Endothelin Receptor Signaling : a Possible Role in the Pathogenesis of Pulmonary Arterial Hypertension"}]},"item_type_id":"3","owner":"11","path":["958"],"pubdate":{"attribute_name":"公開日","attribute_value":"2019-04-10"},"publish_date":"2019-04-10","publish_status":"0","recid":"5521","relation_version_is_last":true,"title":["Involvement of β-arrestin in Endothelin Receptor Signaling : a Possible Role in the Pathogenesis of Pulmonary Arterial Hypertension"],"weko_creator_id":"11","weko_shared_id":-1},"updated":"2023-05-16T05:54:10.677513+00:00"}
